Description

  • The 8F4H7B7 antibody reacts with Vδ6.3/2 T-cell Receptor (TCR)-bearing T lymphocytes
  • Originally defined as a member of the Vδ6 TCR subfamily, it is now proposed that the C57BL-derived Vδ6.3 is an allelic variant of Vδ6.2, found in A/J, AKR, BALB/c, C3H/He, and FVB mice
  • mAb 8F4H7B7 crossreacts with Vδ6.4 and possibly Vδ6.6 in DBA/2 mice, and it also detects a subset ofγδTCR-bearing cells in CBA/J and C57L mice
  • It does not recognize Vδ4, Vδ5, Vδ6.1, or Vδ6.5 TCR
  • A subpopulation of thymocytes expressing Vδ6.3 or Vδ6.4 TCR (in C57BL/6 or DBA/2 mice, respectively) and low levels of CD90.2 (Thy-1.2) shares functional and phenotypic characteristics with NK-T cells
  • SimilarδV 6.4 TCRexpressing lymphocytes make up significant proportions of theγδT-cell populations in the liver and spleen of DBA/2 mice
  • Furthermore, T lymphocytes bearing Vδ6.3/2 TCR are found in the skin and intestinal epithelium and may represent a unique T-cell subpopulation with a potential for autoimmune reactivity
  • This antibody is conjugated to BD Horizon™ BV605 which is part of the BD Horizon Brilliant™ Violet family of dyes
  • With an Ex Max of 407nm and Em Max of 602nm, BD Horizon BV605 can be excited by a violet laser and detected with a standard 610/20nm filter set
  • BD Horizon BV605 is a tandem fluorochrome of BD Horizon BV421 and an acceptor dye with an Em max at 605nm
  • Due to the excitation of the acceptor dye by the green (532 nm) and yellow-green (561 nm) lasers, there will be significant spillover into the PE and BD Horizon PE-CF594 detectors off the green or yellow-green lasers
  • BD Horizon BV605 conjugates are very bright, often exhibiting brightness equivalent to PE conjugates and can be used as a third color off of the violet laser
  • BV605 is a tandem fluorochrome that combines BD Horizon BV421 and Cy™3.5
  • These conjugates are very bright, exhibiting similar brightness to equivalent PE conjugates
  • Due to the excitation of Cy3.5 by the green (532nm) and yellow-green (561nm) lasers, there will be significant spillover of the BD Horizon BV605 signal into the PE and BD Horizon PE-CF594 detectors off the green or yellow-green lasers.
