- TIP60 was initially identified as a protein that interacts with the HIV protein, Tat, and shares no homology with the HATs, CBP/p300, or PCAF
- TIP60 was recently shown to modulate DNA repair and p53 function
- TIP60 also directly acetylatesγ-H2AX and, together with the cofactor, TRRAP, binds to the chromatin surrounding sites of DNA double-stranded breaks in vivo
- Furthermore, TIP60 associates with MYC and recruits it to the chromatin, which results in MYC-induced actetylation of histone H4
- TIP60 directly associates with p53 and specifically acetylates it at K 120 , modulating the decision between cell cycle arrest and apoptosis
- TIP60 acts as a transcription coactivator of p53 that enhances the activation of p21 and puma, leading to cell growth arrest and apoptosis
- K 120 actetylation is essential for p53-mediated puma activation and apoptosis, but is dispensable for both p53-mediated growth arrest, through p21 activation, and p53/MDM2 interaction
- K 120 is not acetylated by p300 and its actetylation by TIP60 does not affect the stability or DNA binding activity of p53.
