  • LIG-1 is a widely expressed single-pass type I membrane protein that acts as a feedback negative regulator of signaling by receptor tyrosine kinases, through a mechanism that involves enhancement of receptor ubiquitination and accelerated intracellular degradation
  • LIG-1 contains 3 Ig-like C2-type (immunoglobulin-like) domains, 15 LRR (leucine-rich) repeats, 1 1 LRRCT domain, and 1LRRNT domain
  • The LRRs and the Ig-domains are each sufficient for EGFR/ERBB1 binding and this interaction is abolished only when these two domains are deleted
  • LIG-1 is highly expressed by intestinal stem cells and controls the size of the intestinal stem-cell niche by regulating the amplitude of growth-factor signaling
  • It is also reported to control stem-cell proliferation in epidermis
