Test Specificity

OCT2 is a transcription factor of the POU homeo-domain family that binds to the Ig gene octamer sites, regulating B-cell-specific genes. OCT2 expression can be used as a marker of B-cell lineage and differentiation. Germinal center B-cells, mantle B-cells, monocytoid B-cells, and plasma cells show high level expression of OCT2. Additionally, mantle cell lymphoma, follicular lymphoma, marginal zone lymphoma, plasmacytoma, Burkitt lymphoma, diffuse large cell lymphoma, diffuse large B-cell lymphoma, Hodgkin lymphoma display increased expression of OCT2. Several studies of OCT2 expression have shown a low level expression in pre-B, T-cell, myelomonocytic, and epithelial cell lines, whereas all mature B-cell lines have high levels of expression. In spite of scanty evidence for OCT2 expression in T cells, it is believed that this factor participates in transcriptional regulation during T-cell activation.

Test Specificity:
Perforin is a pore-forming protein that leads to osmotic lysis of the target cells and subsequently enables granzymes to enter the target cells and activate apoptosis. Perforin has structural and functional similarities to complement component 9 (C9). Like C9, this protein creates transmembrane tubules and is capable of lysing non-specifically a variety of target cells. It is one of the main cytolytic proteins of cytolytic granules, and is known to be a key effector molecule for T-cell- and natural killer-cell-mediated cytolysis. Defects in this gene cause familial hemophagocytic lymphohistiocytosis type 2 (HPLH2), a rare and lethal autosomal recessive disorder of early childhood. The expression of perforin is reportedly upregulated in activated CD8+ T-cells, natural killer cells and some CD4+ T-cells.

Primary or Secondary:

Test Specificity:
Steroidogenic Acute Regulatory Protein (STAR) controls the rate-limiting step of steroidegenesis by translocating cholesterol from the outer mitochondrial membrane to the inner membrane where it is later cleaved to pregnenolone. It is primarily present in steroid-producing cells, including Leydig cells in the testis, theca cells and luteal cells in the ovary and adrenal cells in the adrenal cortex. Due to low levels of pregnenolone, seminomas and Leydig cell tumors display no specific STAR staining. Therefore, STAR antibody may assist in differentiating sex cord stromal tumors (SCST), seminomas and embryonal carcinomas.
