The gp100 molecule is a 100kDa glycosylated protein that is cleaved into a small (26kDa) carboxy-terminal fragment and a larger amino- terminal section (60-64 kDa), which is subsequently cleaved to generate 26kDa and 34-38kDa. By immunohistochemistry, it specifically recognizes a protein in melanocytes and melanomas. This monoclonal antibody reacts with junctional and blue nevus cells and variably with fetal and neonatal melanocytes. Intradermal nevi, normal adult melanocytes, and non-melanocytic cells are negative. It does not stain tumor cells of epithelial, lymphoid, glial, or mesenchymal origin. Metastatic amelanotic melanoma can often be confused with a variety of poorly differentiated carcinomas, large cell lymphomas, and sarcomas using H E stains alone. It is also difficult to differentiate melanoma from spindle cell carcinomas and various types of mesenchymal neoplasms. Recognizes a protein of 150kDa, which is identified as the high molecular weight variant of Caldesmon. Two closely related variants of human caldesmon have been identified which are different in their electrophoretic mobility and cellular distribution. The h-caldesmon variant (120'150kDa) is predominantly expressed in smooth muscle whereas l-caldesmon (70'80kDa) is found in non- muscle tissue and cells. Neither of the two variants has been detected in skeletal muscle. This monoclonal antibody recognizes only the 150kDa variant (h-caldesmon) in Western blots of human aortic media extracts and is unreactive with fibroblast extracts from cultivated human foreskin. Caldesmon is a developmentally regulated protein involved in smooth muscle and non-muscle contraction.

Test Specificity:
This antibody recognizes a protein of 25-26kDa, identified as the Bcl-2 lpha oncoprotein. It shows no cross-reaction with Bcl-x or Bax protein. Expression of Bcl-2 lpha oncoprotein inhibits the programmed cell death (apoptosis). In most follicular lymphomas, neoplastic germinal centers express high levels of Bcl-2 lpha protein, whereas the normal or hyperplastic germinal centers are negative. Consequently, this antibody is valuable when distinguishing between reactive and neoplastic follicular proliferation in lymph node biopsies. It may also be used in distinguishing between those follicular lymphomas that express Bcl-2 protein and the small number in which the neoplastic cells are Bcl-2 negative.
