Description

  • Recombinant rabbit monoclonal antibodies are produced using in vitro expression systems
  • The expression systems are developed by cloning in the specific antibody DNA sequences from immunoreactive rabbits
  • Then, individual clones are screened to select the best candidates for production
  • The advantages of using recombinant rabbit monoclonal antibodies include: better specificity and sensitivity, lot-to-lot consistency, animal origin-free formulations, and broader immunoreactivity to diverse targets due to larger rabbit immune repertoire
  • IKK alpha/beta is encoded by CHUK (conserved helix-loop-helix ubiquitous kinase) and IKKB (inhibitor of kappa light polypeptide gene enhancer in B-cells) gene located in chromosome 10 and 8 respectively in humans
  • Upon stimulation by agents that activate the NF-kB pathway such as TNF alpha, the two subunits of IKK complex, IKK alpha and IKK -beta alongside a regulator NEMO (IKK gamma) phosphorylates IkB proteins
  • This targets the later for ubiquitin mediated degradation and phosphorylation of NF-kB leading to subsequent translocation to the nucleus
  • Phosphorylation at serine 176 and 180 located in the activation loop causes a confirmation change that is required for the activation of the kinases.
