  • Target description: Hypoxia-inducible factor 1-alpha (UniProt: Q16665; also known as HIF1-alpha, HIF-1-alpha, ARNT-interacting protein, Basic-helix-loop-helix-PAS protein MOP1, Class E basic helix-loop-helix protein 78 (bHLHe78), Member of PAS protein 1, PAS domain-containing protein 8) is encoded by the HIF1A (also known as BHLHE78, MOP1, PASD8) gene (Gene ID: 3091) in human. HIF1- is a subunit of the heterodimeric transcription factor HIF-1, which plays a central role in cellular responses to oxygen deficiency. HIF1- has several important domains: an oxygen-dependent degradation domain (ODD) that regulates its stability, a basic helix-loop-helix (bHLH) domain for DNA binding, and Per-ARNT-Sim (PAS) domains for dimerization with HIF1-β. HIF1- is crucial for the activation of genes that respond to hypoxic conditions. Under normal oxygen levels, HIF1- is rapidly degraded by the proteasome system. In hypoxia, it stabilizes and translocates to the nucleus, where it dimerizes with HIF1-β. The HIF-1 complex then binds to hypoxia-responsive elements (HREs) in the DNA, activating genes involved in angiogenesis, metabolism, iron transport, and cell survival. This response is vital for adapting to low oxygen environments and is crucial in processes like embryonic development and wound healing. Its overactivation can contribute to cancer progression by promoting angiogenesis, metabolic reprogramming, and survival under hypoxic tumor microenvironments. Conversely, its activation can be protective in ischemic diseases like myocardial infarction and stroke by promoting adaptive responses. HIF1- is also implicated in chronic kidney disease, where hypoxia is a key factor in disease progression. This ZooMAb® recombinant monoclonal antibody, generated by our proprietary technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals.
  • Physical form: Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS with 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 µL.
  • Reconstitution: 300 µg/mL after reconstitution at 25 µL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.
  • Storage and Stability: Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 µL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
