Description

  • General description: We are committed to bringing you greener alternative products, which adhere to one or more of the 12 Principles of Green Chemistry. This antibody is preservative-free, produced without the harm or sacrifice of animals and exceptionally stable to allow for ambient shipping and storage if needed, and thus aligns with "Waste Prevention", "Designing Safer Chemicals" and "Design for Energy Efficiency". Click here for more information.
  • Specificity: Clone 1B14 is a ZooMAb® rabbit recombinant monoclonal antibody that detects Cullin-1. It targets an epitope within 25 amino acids from the C-terminal region.
  • Immunogen: KLH-conjugated linear peptide corresponding to 25 amino acids from the C-terminal region of human Cullin-1.
  • Application: Quality Control TestingEvaluated by Western Blotting in Ramos cell lysate.Western Blotting Analysis: A 1:1,000 dilution of this antibody detected Cullin-1 protein in Ramos cell lysate.Tested ApplicationsWestern Blotting Analysis: A 1:1,000 dilution from a representative lot detected Cullin-1 in lysates from human testis and mouse testis tissue. Immunohistochemistry (Paraffin) Analysis: A 1:100 dilution from a representative lot detected Cullin-1 in human cerebral cortex and human testis cancer tissue sections.Immunocytochemistry Analysis: A 1:100 dilution from a representative lot detected Cullin-1 in A431 cells.Note: Actual optimal working dilutions must be determined by end user as specimens, and experimental conditions may vary with the end user.
  • Target description: Cullin-1 (UniProt: Q13616; also known as CUL-1) is encoded by the CUL1 gene (Gene ID: 816) in human. Cullin-1 is a core component of the SCF (SKP1-CUL1-F-box protein) E3 ubiquitin ligase complex. It is characterized by a series of cullin repeats that form a long stalk-like structure, with a C-terminal domain that interacts with the RING protein RBX1, and an N-terminal domain that interacts with SKP1 and various F-box proteins. This scaffold-like structure is crucial for assembling the other components of the SCF complex, facilitating the ubiquitination of target proteins. CUL1′s primary function is to mediate the ubiquitination and subsequent proteasomal degradation of various proteins involved in cell cycle regulation, signal transduction, and transcription. Through the SCF complex, CUL1 contributes to the regulation of processes such as cell division, growth, and response to stress. The specificity of the SCF complex for different substrates is determined by the F-box protein, which varies and binds specific target proteins for ubiquitination. CUL1 is ubiquitously expressed across different tissues, reflecting its fundamental role in cell cycle and growth control. Its expression levels can vary depending on the cell cycle phase and cellular needs, with mechanisms in place to ensure its availability synchronizes with the requirements for substrate degradation. Dysregulation of CUL1 and its associated complex components can lead to diseases characterized by abnormal cell growth and division. This ZooMAb® recombinant monoclonal antibody, generated by our proprietary technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals.
  • Physical form: Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS with 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 µL.
  • Reconstitution: 300 µg/mL after reconstitution at 25 µL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.
  • Storage and Stability: Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 µL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
