• Most clinical specimens received for isolation of acid-fast bacilli are contaminated with more rapidly growing commensal microbial flora
• Respiratory specimens also often contain mucin which may trap the acid-fast bacilli and require liquefaction to release the mycobacterial cells prior to inoculation of media3
