Description

  • Description Kit provides everything needed to covalently label proteins with a dibenzylcyclooctyne (DBCO) group for copper-free click chemistry applications Contains all necessary materials to perform five labeling reactions and subsequent purification for removal of excess DBCO reagent TFP-PEG4-DBCO is a heterobifunctional crosslinker which contains an amine reactive tetrafluorophenyl (TFP), and an azide reactive dibenzylcyclooctyne (DBCO) with a water solubilizing PEG4 spacer in between It readily reacts with the primary amines found on proteins to form a covalently attached DBCO group, which can be further reacted with an azide labeled coupling partner (supplied separately) to create diverse bioconjugates In the first conjugation step, the TFP group reacts with a primary amine on a protein to form a stable amide bond and a covalently bound DBCO moiety Subsequent purification removes excess crosslinker from solution which may interfere with the second labeling step Once a protein is DBCO labeled and purified, it can undergo a copper free strain-promoted azide-alkyne cycloaddition (SPAAC) Label proteins (20 to 150 kDa) with concentrations ranging from 0.5 to 5 mL TFP esters are more hydrolytically stable than NHS esters enabling less reagent to be used Labeling and purification can be completed in as little as 90 minutes with 30 min hands-on time
