  • Target description: Protein RUFY3 (UniProt: Q7L099; also known as RUN and FYVE domain-containing protein 3, Rap2-interacting protein x, RIPx, Single axon-regulated protein, Singar) is encoded by the RUFY3 (also known as KIAA0871) gene (Gene ID: 22902) in human. RUFY3 is a RUN domain-containing protein that plays a role in the generation of neuronal polarity formation and axon growth. It is implicated in the formation of a single axon by developing neurons. It may inhibit the formation of additional axons by inhibiting PI-3 kinase in minor neuronal processes. It is reported to specifically interact with actin filament-binding proteins, such as Fascin, and colocalizes with Fascin in growth cones. Knockdown of RUFY3 gene impairs the distribution of Fascin and actin filaments, accompanied by an increased proportion of neurons with multiple axons and a decrease in the axon length. RUFY3 also serves as an adapter protein for small GTPase proteins and is bound to activated Rap2, a Ras family protein in the developing neurons. Overexpression of RUFY3 leads to the formation of F-actin-enriched protrusive structures at the cell periphery and induces gastric cancer cell migration. It is reported that p21-activated kinase-1 (PAK1) interacts with RUFY3 and promotes its expression. Inhibition of PAK1 attenuates RUFY3-incuded cell migration and invasion. This ZooMAbZooMAb® recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals.   • Physical form: Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS, 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 μL.
  • Reconstitution: 30 μg/mL after reconstitution at 25 μL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.
  • Storage and Stability: Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 μL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
