  • Target description: Myosin-10 (UniProt: P35580; also known as Cellular myosin heavy chain, type B, Myosin heavy chain 10, Myosin heavy chain, non-muscle IIb, Non-muscle myosin heavy chain B, NMMHC-B, Non-muscle myosin heavy chain IIb, NMMHC II-b, NMMHC-IIB) is encoded by the MYH10 gene (Gene ID: 4628) in human. Myosins are highly conserved, ubiquitously expressed proteins that form regular bipolar thick filaments that together with actin thin filaments constitute the fundamental contractile unit of skeletal and cardiac muscle. Myosin-10 is a member of the MyTH4/FERM domain family of unconventional, actin-based motor proteins. It contains two heavy chain subunits (MHC), two alkali light chain subunits (MLC) and 2 regulatory light chain subunits (MLC-2). It has a rodlike tail sequence that is highly repetitive, showing cycles of a 28-residue repeat pattern composed of four heptapeptides, characteristic for -helical coiled coils. It is mainly localized to cell projections and lamellipodia. Myosin-10 plays a role in cytokinesis, cell shape, and specialized functions such as secretion and capping. During cell spreading, it plays an important role in cytoskeleton reorganization, focal contacts formation, and lamellipodial extension. Myosin-10 is reported to drive glioblastoma growth by stimulating integrin-dependent signaling. Its levels are upregulated in lung, prostate, and breast carcinomas. Myosin-10 knockout mice can survive to adulthood, implying that normal cells can compensate for its loss. Its deletion is shown to impair invasion, slow proliferation, and prolongs survival in murine models of glioblastoma. Five isoforms of Myosin-10 have been described that are produced by alternative splicing. This ZooMAb® recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals.   • Physical form: Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS with 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 μL.
  • Reconstitution: 30 μg/mL after reconstitution at 25 μL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.
  • Storage and Stability: Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 μL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
