  • Target description: T-cell surface glycoprotein CD8 alpha chain (UniProt: P01732; also known as T-lymphocyte differentiation antigen T8/Leu-2, CD8a) is encoded by the CD8A (also known as MAL) gene (Gene ID: 925) in human. CD8 alpha is a single-pass type I membrane glycoprotein that is synthesized with a signal peptide (aa 1-21), which is subsequently cleaved to produce the mature protein that has an extracellular domain (aa 22-182), a helical domain (aa 183-203), and a cytoplasmic tail (aa 204-235). CD8 is generally present as a heterodimer of and β chains linked by two disulfide bonds, however, it can also form homodimers. The CD8 / homodimer is expressed on a subset of / TCR-bearing T cells, NK cells, intestinal intraepithelial lymphocytes, and lymphoid dendritic cells. CD8a homodimer is reported to promote the survival and differentiation of activated lymphocytes into memory CD8 T-cells. CD8a plays an essential role in the immune response and serves multiple functions in responses against both external and internal offenses. In T-cells, it functions primarily as a co-receptor for MHC class I molecule:peptide complex. It interacts simultaneously with the T-cell receptor (TCR) and the MHC class I proteins presented by antigen presenting cells. In turn, it recruits the Src kinase LCK to the vicinity of the TCR-CD3 complex where LCK phosphorylates various substrates leading to lymphokine production, motility, adhesion, and activation of cytotoxic T-lymphocytes (CTLs). This process enables CTLs to recognize and eliminate infected cells and tumor cells. Mutations in CD8A gene are known to cause the absence of CD8+ cells leading to recurrent bacterial infections. This ZooMAb® recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals.   • Physical form: Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS with 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 µL.
  • Reconstitution: 300 µg/mL after reconstitution at 25 µL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.
  • Storage and Stability: Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 µL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
