Description

  • Description: E-Selectin (CL-37) is a mouse monoclonal antibody raised against E-Selectinof human origin
  • Endothelial Leukocyte Adhesion Molecule-1 belongs to the selectin family of adhesion molecules
  • Together with L-selectin and P-selectin, E-selectin mediates the initial interactions of leukocytes and platelets with endothelial cells
  • Molecular structure: The extracellular part of all selectins consists of an aminoterminal c-type lectin domain which specifically binds to carbohydrate ligands
  • This is followed by an EGF-like domain, and, in the case of E-selectin, by 6 short consensus repeats
  • The transmembrane portion of the molecule is followed by a short cytoplasmic tail
  • Selectins guide non-activated polymorphonuclear cells to the areas of inflammation in creating first, loose contacts with the endothelial layer
  • Together with P-selectin, E-selectin is expressed on cytokine-activated endothelial cells, and contributes to the adhesion of still resting leukocytes to the endothelium
  • This initial binding event is an essential prerequisite for the activation of the immune cells via different inflammatory mediators
  • In contrast to P-selectin, E-selectin is maximally expressed 2-4 hours after cell activation
  • Within the next 24-48 hours E-selectin is again eliminated from the cytoplasmic membrane by shedding into the circulation

  • Leukocytes adhere to the blood vessel endothelium during extravasation in postcapillary venules of lymph nodes
  • In addition, leukocyte adhesion occurs in the capillaries and small venules at any site in the body after onset of inflammation
  • This response is immediate and involves a cascade of adhesion receptors
  • At the endothelial surface members of the selectin and immunoglobulin superfamilies participate in this cascade
  • Selectins are C-type cell surface lectins that play a role in leukocyte adhesion to the blood vessel wall endothelium
  • E-selectin (CD62E) is an endothelial cell specific selectin that is expressed only after activation with proinflammatory cytokines
  • In vitro experiments have shown that IL-1, TNFalpha and bacterial wall components like lipopolysaccharides induce the transcription of E-selectin in a NFkB dependent signaling cascade
  • E-selectin has been associated with blood vessel endothelium in diverse inflammatory situations.
