- Specifically detects Clathrin Heavy Chain Clone: X22 in Bovine, Human, Mouse, Rat samples, and it is validated for Immunofluorescence, Immunoprecipitation At the unstimulated presynaptic nerve terminal, a pool of synaptic vesicles remains docked at active zones in preparation for exocytosis
- Nerve terminal depolarization induced through an action potential stimulates the rise in intracellular Ca 2+ , which ultimately initiates the exocytotic process
- Fused synaptic vesicles are rapidly replaced with vesicles from the reserve pool, while membranes of the fused vesicles are recaptured by endocytosis
- Clathrin is one component involved in vesicle endocytosis, and is composed of three polypeptides, a ∽180kDa heavy chain and two ∽32-38kDa light chains which combine to create a distinct three-legged triskelion
- It is this morphology that allows clathrin to form its unique polyhedral network
- The clathrin-adaptor AP2 and the AP180/AP3 proteins also play a role in this process by forming a protein layer between the membrane and clathrin lattice
- The interaction of the AP2 heterotetramer with synaptotagmin is believed to act as the template for recruitment of the AP3 and clathrin triskelia complex
- Progression from clathrin-coated to free vesicles in the nerve terminus also requires dynamin.
