Test Specificity

This MAb recognizes a protein of 56.5 kDa, identified as cytokeratin 10 (CK10). CK10 is expressed in all suprabasal layers of the epidermis. In the epidermis, expression of CK10 strictly parallels the extent of differentiation; it is absent in the basal layer, appears in the first suprabasal layers and increases in concentration towards the granular layer. However, CK10 is rarely detected in early stages of vulvar squamous carcinomas (tumors less than 2 cm, clinical stage I) regardless of the tumor grade. In larger and more advanced tumors (greater than 2 cm, clinical stages II and III), CK10 is detected very frequently. Expression of CK10 is related to maturation of malignant keratinocytes, being preferentially detected in more-differentiated parts.

Elastin is a polymeric protein and is the main component of the extracellular matrix of arteries. It is synthesized and secreted as a soluble, single-chain protein (tropoelastin), which undergoes a number of post-translational modifications prior to its organization into an elastic fiber in the extracellular space. Elastin performs a regulatory function during arterial development by controlling proliferation of smooth muscle and stabilizing arterial structure. It imparts elasticity to the connective tissue. During aging, the elasticity of connective tissue is reduced because of the cross-linking of collagenous fibers with elastin. The abnormal accumulation of elastic tissue in blood vessels is found in atherosclerosis and hypertension.

Recognizes a phospho-protein of 45kDa, identified as MyoD1. This monoclonal antibody does not cross react with myogenin, Myf5, or Myf6. Antibody to MyoD1 labels the nuclei of myoblasts in developing muscle tissues. MyoD1 is not detected in normal adult tissue, but is highly expressed in the tumor cell nuclei of rhabdomyosarcomas. Occasionally nuclear expression of MyoD1 is seen in ectomesenchymoma and a subset of Wilm s tumors. Filaggrin is an intermediate filament-associated protein that aggregates keratin intermediate filaments in mammalian epidermis. It is initially synthesized as a polyprotein precursor, profilaggrin (consisting of multiple filaggrin units of 324 aa each), which is localized in keratohyalin granules, and is subsequently proteolytically processed into individual functional filaggrin molecules. Active filaggrin is present at a level of the epidermis where keratinocytes are in transition between the live nucleated granular layer and the anucleate cornified layer, suggesting that filaggrin aids in the terminal differentiation process by facilitating apoptotic machinery.
