Description

  • General description: We are committed to bringing you greener alternative products, which adhere to one or more of the 12 Principles of Green Chemistry. This antibody is preservative-free, produced without the harm or sacrifice of animals and exceptionally stable to allow for ambient shipping and storage if needed, and thus aligns with "Waste Prevention", "Designing Safer Chemicals" and "Design for Energy Efficiency". Click here for more information.
  • Specificity: Clone 3H3 is a ZooMAb® recombinant rabbit monoclonal antibody that specifically detects Thioredoxin/TXN. It targets an epitope within the full length protein.
  • Immunogen: His-tagged, Full length human Thioredoxin/TXN recombinant protein.
  • Application: Quality Control TestingEvaluated by Western Blotting in MCF7 cell lysate.Western Blotting Analysis: A 1:1,000 dilution of this antibody detected Thioredoxin/TXN in MCF7 cell lysate.Tested ApplicationsWestern Blotting Analysis: A 1:1,000 dilution from a representative lot detected Thioredoxin/TXN in HepG2, NIH/3T3 cell lysates, and human small intestine tissue lysate.Immunohistochemistry (Paraffin) Analysis: A 1:1,000 dilution from a representative lot detected Thioredoxin/TXN in human esophagus.Flow Cytometry Analysis: 0.1 µg from a representative lot detected Thioredoxin/TXN in one million HepG2 cells.Immunocytochemistry Analysis: A 1:100 dilution from a representative lot detected Thioredoxin/TXN in HepG2 cells.Note: Actual optimal working dilutions must be determined by end user as specimens, and experimental conditions may vary with the end user.
  • Target description: Thioredoxin (UniProt: P10599, also known as ATL-derived factor (ADF) and surface-associated sulphydryl protein (SASP) is encoded by TXN (also known as TRX1) gene (Gene ID; 7295) in human. Thioredoxin is a small protein with a conserved active site sequence containing two cysteine residues, which are present in many organisms. Impaired cellular redox balance has been linked with age-related diseases, and neurodegenerative diseases, such as Alzheimer and Parkinson′s disease. Thioredoxin 1 contains two cysteine amino acid residues in a vicinal arrangement (i.e., next to each other), which allows it to form an internal disulfide bond, or disulfide bonds with other proteins. As such, it can be used as a repository of reduced or oxidized disulfide bond moieties. As its name would imply, Thioredoxin 1 functions to reduce oxidative stress within cells and thus promotes better health. Interestingly mice that over express Thioredoxin 1 appear to live much longer and are more resistant to inflammation than nonover expressing mice suggesting that thioredoxin s efforts in reducing the number of free radicals and other oxidizing agents within cells leads to longevity. Thioredoxin 1 plays a role in nitric oxide metabolism, and can act as an active inhibitor of caspase 3 by nitrosylating caspase 3 s active site in response to NO activation. Thioredoxin also induces AP-1 protein and FOS/JUN gene expression after ionizing radiation, thereby providing a mechanism aimed at cell survival. This ZooMAb® recombinant monoclonal antibody, generated by our proprietary technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals.
  • Physical form: Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS with 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 µL.
  • Reconstitution: 300 µg/mL after reconstitution at 25 µL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.
  • Storage and Stability: Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 µL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
