  • coli cells are ideal for high-efficiency cloning and plasmid propagation, achieving a transformation efficiency of >1 x 10 8 cfu/μg with control plasmid DNA in StripWell format
  • These cloning competent cells are packaged in a rack containing 12 strips of 8 tubes to increase productivity for throughput bacterial transformations

  • coli cells are the laboratory workhorse for chemical transformations as they are compatible with a broad spectrum of DNA types for transformation
  • They allow stable replication of high-copy number plasmids and are the same competent cells that come with many of our cloning kits
  • TOP10 E
  • coli cells are genetically similar to the DH10B strain and have been reported to be more resilient to stress conditions (osmotic shock and acidic pH stress)
  • These cells carry mutations in the methylation-dependent restriction system ( mcr A, mcr BC, and mrr ), allowing cloning of both prokaryotic and eukaryotic genomic DNA, as well as efficient plasmid rescue from eukaryotic genomes
  • Similar to other DH strains, TOP10 cells contain rec A1 and end A1 mutations that increase insert stability and improve the quality of plasmid DNA prepared from minipreps

  • coli cells offer: • Transformation efficiencies of >1 x 10 8 cfu/μg • hsd R for efficient transformation of unmethylated DNA from PCR amplifications • mcr A for efficient transformation of methylated DNA from genomic preparations • lac ZΔM15 for blue/white color screening of recombinant clones on plates containing either X-Gal or Bluo-Gal • end A1 for cleaner preparations of DNA and better results in downstream applications due to the elimination of non-specific digestion by endonuclease I • rec A1 for reduced occurrence of non-specific recombination in cloned DNA Note: TOP10 cells do not require IPTG to induce expression from the lac promoter
  • MultiShot StripWell flexible format Each StripWell of eight connected tubes is designed with the same single-tube, single-use features as our One Shot format tubes
  • This format allows all steps of the transformation protocol, up to plating, to take place in the same tube, thereby saving time and reducing chance of contamination
  • After addition of DNA, MultiShot StripWell competent cells can be transformed by heat-shock at 42°C using a water bath, all in the same tube
  • StripWell tubes can be cut to permit transformations in a single tube, eliminating wasted aliquots and avoiding freeze-thaws that can result in reduced transformation efficiency
