  • Target description: UL16-binding protein 1 (UniProt: Q9BZM6; also known as ALCAN-beta, NKG2D ligand 1, N2DL-1, NKG2DL1, Retinoic acid early transcript 1I) is encoded by the ULBP1 (also known as N2DL1, RAET1I) gene (Gene ID: 80329) in human. The UL16-binding proteins are a novel family of MHC class I-related, GPI-anchored, cell surface proteins that lack an 3 domain, and do not associate with β2-microglobulin. ULBP1 was first identified based on its ability to bind to the human CMV (HCMV) glycoprotein UL16. It is synthesized with a signal peptide (aa 1-25) and a propeptide (aa 217-244) that are subsequently cleaved off to produce the mature protein (aa 26-216). It is expressed in T-cells, B-cells, erythroleukemia cell lines and in a wide range of tissues. It is also found in fetal heart, brain, lung, and liver. It binds and activates the natural killer group 2 member D (NKG2D) receptor and mediates natural killer cell cytotoxicity. In CMV-infected cells, it interacts with the viral glycoprotein UL16 and this interaction causes ULBP1 retention in the endoplasmic reticulum and cis-Golgi and prevents binding to and activation of NKG2D, which provides CMV with an immune evasion mechanism. ULBP1 expressed on the tumor cell surface can also bind to the NKG2D receptor presenting on NK cells and CD8+ T and T cells. Patients with NKG2D expression in gastric tumor exhibit significantly longer overall survival compared to those without NKG2D expression in tumors. This ZooMAb® recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals.   • Physical form: Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS, 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 μL.
  • Reconstitution: 300 μg/mL after reconstitution at 25 μL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.
  • Storage and Stability: Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 μL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
