Differentiate genera within the Enterobacteriaceae on the basis of urease production with Thermo Scientific™ Remel™ Urea Broth (Stuart's)
Urea Broth (Stuart's) was developed by Stuart et al
to differentiate Proteus from other genera within the Enterobacteriaceae 1,2
This medium is designed such that it detects urease activity in organisms which produce ammonia in a highly buffered medium with limited nutrients
Such organisms include Proteus spp., Morganella morganii , Providencia rettgeri , and urease-positive Providencia stuartii 3
Urea Broth (Stuart’s) is a liquid medium used to detect microorganisms on the basis of urease production
It is a medium with high buffering capacity and limited nutrients
Other genera within the Enterobacteriaceae which hydrolyze urea, will not do so in this medium
Ready to use – Liquid medium
Easy to interpret – An intense pink-red color throughout the broth indicates a positive test
No color change, the broth remains yellow-orange in the case of a negative test
This medium contains yeast extract that provides essential growth factors required by strong urease-positive organims such as Proteus spp., M
morganii , and some species of Providencia
Urea is the substrate that can be hydrolyzed
Disodium and monopotassium phosphates provide the buffering system
Phenol red acts as the pH indicator
Organisms which possess the urease enzyme hydrolyze urea and produce an alkaline by-product (ammonia) which is detected when the phenol red indicator changes color from yellow-orange to pink
