Description

  • Selectively recover and differentiate Salmonella spp
  • other than Salmonella Typhi with Thermo Scientific™ Remel™ XLT4/ Brilliant Green w/Novobiocin Biplate
  • XLT4 (Xylose-Lysine-Tergitol 4) Agar was developed by Miller and Tate to selectively screen samples contaminated with enteric organisms for the presence of Salmonella 1
  • They found that XLT4 Agar completely inhibits competing enteric organisms
  • Brilliant Green Agar was developed by Kristensen et al
  • to isolate Salmonella 2
  • Novobiocin was added to Brilliant Green Agar to supress the growth of competing organisms and to improve selectivity 3,4
  • Salmonella infection (salmonellosis) is a common bacterial disease that affects the intestinal tract
  • Salmonella bacteria typically live in animal and human intestines and are shed through feces
  • Humans become infected most frequently through contaminated water or food
  • Salmonella is a rod-shaped, gram-negative facultative anaerobe that belongs to the family Enterobacteriaceae and is one of the most commonly isolated foodborne pathogens found predominantly in poultry, eggs and dairy products
  • Numerous Salmonella serotypes have been identified, the majority of which belong to Salmonella enterica subsp
  • enterica and can cause serious, at times life-threatening, infections in humans
  • The Biplate contains XLT Agar in one section and Brilliant Green Agar with Novobiocin in the other section
  • This convenient biplate format produces more diagnostic details of pathogenic organisms while reducing storage capacity issue in incubators and allows simultaneous detection of sensitive pathogens, providing a complete diagnostic picture for fast decision making
  • XLT4 Agar is a solid medium that is used for selective and differential recovery of non-typhi Salmonella species whereas Brilliant green agar with novobiocin allows for selective recovery of Salmonella spp
  • other than serovars Typhi and Paratyphi
  • Lactose and sucrose fermenting colonies occasionally growing on Brilliant Green Agar medium will be yellow-green in color surrounded by a yellow-green zone
  • Salmonella colonies range from reddish or pink to nearly white in color with a red zone
  • Easy-to-use – Convenience of prepared media Space saving biplate format – Allows simultaneous detection of sensitive pathogens Selective media –Tergitol 4 reduces the growth of non-salmonellae enteric gram-negative bacilli in XLT4 Agar
  • Brilliant green dye in the Brilliant green agar inhibits the growth of non-target organisms
  • Novobiocin suppresses the growth of Proteus and, to a lesser extent, Escherichia coli , Citrobacter , and Pseudomonas Easy to interpret – Salmonella forms reddish or pink to nearly white with a red zone colonies in Brilliant Green Agar while forming black centered colonies in XLT4 agar Section I of the biplate consists of XLT4 agar that contains meat peptone and yeast extract that supply nitrogenous compounds, amino acids and vitamins necessary for bacterial growth and xylose as the carbohydrate source
  • Lysine helps to isolate salmonellae based on their ability to decarboxylate lysine and produce an alkaline pH
  • Lactose and sucrose are added in excess to prevent colonies of lysine-positive coliforms from reverting to alkaline conditions
  • Sodium thiosulfate is a sulfur source and ferric ammonium citrate is an indicator of H 2 S-production
  • Tergitol 4 acts as a selective agent and enhances the specificity and sensitivity of Salmonella detection and reduces the growth of non-salmonellae enteric gram-negative bacilli
  • Section II contains Brilliant Green Agar with Novobiocin which has casein and meat peptones to provide nitrogen, amino acids and peptides necessary for bacterial growth
  • Lactose and sucrose are the fermentable carbohydrates
  • Yeast is a source of B-complex vitamins, sodium chloride provides essential electrolytes and maintains osmotic equilibrium and brilliant green dye is the selective agent that inhibits gram-positive bacteria and most gram-negative bacilli
  • Novobiocin is the antibiotic that supresses growth of competing organisms
  • Phenol red is the indicator
