Immunogen

Galiximab is a primatized mAb which consists of human constant and primate (cynomolgus macaque) variable regions and binds specifically to B7-1/CD80. B7-1/CD80 is a surface glycoprotein and a member of the B7 family of costimulatory molecules. B7-1/CD80 antigen regulates T cell activation (through interacting with CD28 or CD152) and is expressed transiently in antigen-presenting cells, T cells and normal B cells, and expressed consitutively on various subtypes of B-cell lymphomas.

Test Specificity:
A54 binds specifically to the D1 domain of CD40/TNFRSF5 (and can only bind to the intact i.e. non-truncated version of CD40/TNFRSF5 and when an AD2 peptide tag has not been added) and can recognize CD40/TNFRSF5 on a B cell line (BJAB). This scFv binds to CD40/TNFRSF5 (immobilized by amine coupling) with a KD within the range of 40nM. CD40/TNFRSF5 is a glycoprotein of the TNFR superfamily and is expressed on all mature B cells, dendritic cells, activated monocytes, some endothelial cells and some epithelium including the thymus. CD40/TNFRSF5 binds to the membrane protein CD154 of the TNF superfamily and this interaction is required for the seconfary immune responses and germinal centre formation.

3F12 binds to human LTalpha3 and LT-alpha-beta (with a KD of ∽0.3 nM, determined by BIACORE; ∽ 37 pM, determined by ELISA) in both its soluble homotrimeric and membrane heterotrimeric forms. LTalpha belongs to the tumor necrosis factor (TNF) superfamily and is secreted as a homo-trimer (LTalpha3), or is expressed on the cell surface in complex with LTbeta.
