- Signal 1-34, Membrane cofactor protein (MCP) or CD46 is a widely expressed complement regulator that inhibits all three pathways of complement activation, classical (CP), alternative (AP), and lectin (LP)
- It binds to both C3b (AP) and C4b (CP) and acts as a cofactor for their inactivation by the C3b/C4b inactivator enzyme factor I
- CD46 is initially produced with an N-terminal signal sequence (a.a
- 1-34), which is post-translationally removed to yield the type 1 membrane bound CD46 consisting of a large extracellular (a.a
- 35-343) portion, followed by a single transmembrane α-helical region (a.a
- 344-366) and a cytoplasmic tail (a.a
- 367-392) with several variant structures
- The extracellular portion is composed of four extracellular Complement Control Protein (CCP) domains carrying the cofactor activity, a collar STP-rich region, and a short region of unknown function (U)
- CD46 also acts as a receptor for certain pathogens such as the measles virus (Edmonston strain), human herpesvirus 6, and Neisseria gonorrhoeae
- CD46 is also implicated to have a role in human egg fertilization
- Several mutations in the CCPs and the transmembrane region of CD46 are linked to atypical hemolytic uremic syndrome (aHUS).
