- Ligand binding to a variety of cell surface receptors, like cytokine and growth factor receptors, results in the association and subsequent activation of JAK kinases via phosphorylation
- Activated JAK proteins, in turn, phosphorylate members of the Signal Transducers and Activators of Transcription (STAT) family of transcription factors
- In the JAK2 domain, there is a region that has considerable sequence homology to the regulatory region of the insulin receptor and contains two tyrosines, Tyr 1007 and Tyr 1008 , that are key phoshorylation/regulatory sites.
